Summary The influence of 4 opioid receptor agonists on Ca2+ uptake by the bovine aortic media was investigated in vitro. 45Ca was used and radioactivity was counted in a beta scintillation counter. Morphine 5pM and morphiceptin 5pM, both p-opioid receptor agonists with a very low affinity for 5-opioid binding sites, have a negligible effect on Ca3+ uptake by the preparation. Dynorphin 1-13, 5 μM, agonist of K-opioid receptors, increases Ca2+ uptake, but this action is not inhibited by naloxone 500 nM, an opioid receptor antagonist. Methionin-5-enkephalin 5 μM, a 6-opioid receptor agonist, increases Ca2+ uptake ranging from 10-12%, this action is constant and fully antagonized by naloxone 500 nM. Endothelium exerts an inhibiting action on Ca2+ uptake by the preparation and this action is diminished in the presence of methionin-5-enkephalin 5 μM. These results indicate the existence of 6-opioid receptors on the examined vascular smooth muscle: the stimulation of which results in an increase of Ca2+ uptake by the preparation.
