| Summary | Studying the effects of pro-opiomelanocortin (POMC) peptides on adrenocortical steroidogenesis we showed that α-MSH has a specific glomerulotropic effect. The effects of synthetic ACTH-(4-7), ACTH-(6-10), ACTH-(7-10), ACTH-(4-10) and ACTH-(11-13) were studied on aldosterone production of isolated zona glomerulosa cells and on the corticosterone production of isolated zona fasciculata cells. With the exception of ACTH-(7-10) all tested fragments had a steroidogenic effect on both type of cells. ACTH-(4- 10)-NH2, Ac-ACTH-(4-10) and Ac-ACTH-(4-10)- NH2, increased zona glomerulosa aldosterone-, but had no effect on zona fasciculata corticosterone production. Studying the effect of Ala-ACTH-(4-10) analogues we found that the effect of Ala5– ACTH-(4-10) was similar to the original compound, while the effect of Ala4, Ala8 and Ala9 analogues at 10-3 M concentration was 5-10% of the effect of ACTH-(4-10). β-endorphin enhances, inhibits or has no effect on corticosteroidogenesis, depending on the dose and on the functional state of the adrenocortical cells (saturation of receptors). It is suggested that the modulatory effects of POMC peptides on adrenocortical steroidogenesis may be of physiological significance. | |
