| Summary | Blockade of the renin-angiotensin system is of primary importance in the treatment of hypertension and congestive heart failure. A breakthrough came with the development of angiontensin converting enzyme (ACE) inhibitors that suppress angiotensin biosynthesis. A more selective approach has now been realised through the discovery of imidazole-based non-peptide angiotensin II (ANGII) receptor antagonists. Lead compounds in this series of novel therapeutics agents all maintain the same substitution pattern around the imidazole nucleus. To investigate the effect of changing the orientation of the imidazole ring on biological activity, a series of analogues was prepared and evaluated as ANGII receptor antagonists in vitro, and as antihypertensive agents in vivo. The findings of this study will be discussed in relation to developments within this field. | |
