Summary | During the second phase of our research work we wanted to test different yeast strains of the Culture Collection of the Institute of Chemistry (MZKI) for their ability to produce HMG CoA reductase inhibitors. After growing those on beer wort and potato dextrose agar slants the yeasts were cultivated in a two-step fermentation procedure in a substrate sustaining the production of inhibitors (1). The vegetative phase of the fermentation lasted 2 days and the production 1-5 days. AH experiments were carried out on a rotary shaker at 30 °C (200 rpm). After fermentation, the fermentation broth was extracted with methanol, filtered, converted to a sodium salt and lactonized (2). The extracts were initially analysed by TZC (3), and followed by HPLC analyses (4). Lovastatin was used as a standard in a tablet formulation (Mevacor -Merck). The isolated compounds were further analysed by mass spectrometry and the spectra compared with the standard sample of lovastatin. The isolated com-pounds of the two strains of Candida cariosilignicola and Pichia labacensis were tested also by means of a biological test using HMG CoA reductase from CHO-K1 cells (5,6). | |
