Summary The effect of mesulergine a 5-hydroxytryptamine (5-HT) receptor antagonist with dopaminergic properties on feeding behavior and brain serotonin and dopamine turnover was investigated. Male Wistar rats were i.p. injected 1 and 3 mg/Kg mesulergine (9.30 a.m.) and allowed free access to food from two feeders with standard rats diet (SD), and carbohydrate enriched diet (CED) on food deprivation schedule. After last mesulergine dose animals were decapitated for serotonin and dopamine determinations in brain areas, according to HPLC method. There was a significant increase of SD and CED intake at 4 hrs with increase in the number of meals and meal size only for the SD while feeding parameters of the CED were not significantly changed. At 24 hrs SD intake was still increased with concomitant increase of the number of meats and rate of feeding while CED was reduced with a decrease of the rate of feeding and meal size. An enhanced serotonergic activity in hippocampus with a decreased dopaminergic activity in hypothalamus and striatum was observed. The data suggest that dietary choices induced by mesulergine are mediated by 5-HT2q and D2 receptor sites in brain.
