Summary | Phototoxicity or photoirritation has been evaluated in rats and guinea pigs by various methods as a safety test prior to initial exposure of human. These methods, besides the fact that they are expensive, require killing of lots of animals, which is unwanted. We have modified an assay for evaluation of medicines for phototoxic potential prior to human testing. Our assay uses Saccharomyces cerevisiae, seeded in an agar overlay on top of a plate count agar base. Activity is quantified by cytotoxicity measured as zone of inhibition. We detect photobiological activity for a big number of medicines. As positive control we use 8-mejhoxypsoralen (8-MOP) at a concentration of 10-2 %. The creation of inhibition clear zones of the Saccharomyces cerevisiae development suggest the phototoxic results of the substance we are studying. | |
