Summary This review outlines the major classes of environmental estrogens, their source and their effects on human’s health. The mechanism of action of selective estrogen receptor modulators (SERM’s), mediated mainly by the intracellular estrogen receptors is discussed thoroughly. The alpha and beta estrogen receptor types and the exon deleted forms, their structure-function relationship and their interaction with DNA and/or other regulatory molecules is described. The role of SERM’s in the selective modulation of the estrogen receptor and the significance of tissue specific proteins in modulating the estrogenic or antiestrogenic activity on the tissue is further discussed. The effect of estrogens, SERMs and phytoestrogens on breast and endometrial cancer, on atherosclerosis and coronary disease, on autoimmune diseases, on osteoporosis and on Alzheimer’s disease is also mentioned.
