Summary TNF-α is a cytokine with a key role in the control of body mass in normal weight-controlled situations while abnormalities in either its production during cachexia or its action during obesity are responsible for the lack of control of body weight. Sibutramine is a noradrenaline and serotonin reuptake inhibitor, which induces weight loss by enhancing satiety and increasing thermogenesis. The aim of the study was to investigate if eventual changes on High Fat Diet intake (HFD) and Body Weight caused by (S) could be correlated to alterations on serum TNF-α levels of rats. 42 male Wistar rats separated in 2 Groups (n=21/ Group) were fed with HFD or Standard Diet (SD) for 13 weeks. The last 3 weeks each Group divided into 3 sub- groups (n=7/subgroup) received: Sibutramine-HCL (S) 5mg/kg, (S) 10 mg/kg or vehicle. Food intake was measured daily the last week of the experiment, while Body Weight weekly. Caloric intake and Body Weight-Increasing Rate (BW-IR) were calculated and serum TNF-α levels were estimated by Elisa. HFD increased caloric intake and BW-IR of rats compared to SD. A significant decrease on food intake and BW-IR of rats fed the HFD after (S) treatment at 10 mg/kg compared to vehicle treatment was observed. Serum TNF-α levels were increased in all rats fed the HFD compared to those fed the SD. Results suggest that HFD intake increases TNF-α levels in serum, or this is the result of BW-IR of rats after fat intake. Sibutramine, by its actions on appetite and thermogenesis, tries to reverse this situation without involvement of TNF-α on its effects.
