Summary The human hair follicle cycle is nowadays adequately understood. The molecular mechanisms responsible for hair growth that involve a cascade of genes, is also being unraveled by a series of studies. The human hair follicle represents a unique, highly regenerative neuroectodermal-mesodermal interaction system that contains numerous stem cells. As the stem cell technology expands to the understanding and therapy of conditions such as cancer and ageing the exploitation of the human follicle cycle is of ever more importance. Finally, the melanocortins involved in the growth stimulation of follicular stem cells, may contribute to the better understanding of cytoprotection against carcinogens. This article gives an adequate knowledge about the structural biology of the hair follicle. A discussion about hormonal, neural and growth factors involved is also given. Moreover, the nature of the stem cells responsible for the hair follicle cycling is being exploited together with the role of α-MSH, a melanocortin that has pleiotropic actions in human skin cells. Finally, the management of common hair growth disorders and the potent modulators of human hair follicle cycling are stated.
