Summary Signal Transducer and Activator of Transcription (STAT) proteins are transcription factors known to be activated by G protein-coupled receptors including opioid receptors. Recent observations have shown that STAT5A interacts directly with the C-terminal region of the μ-opioid receptor (μ-OR) and is phosphorylated upon receptor stimulation. In this regard, we aimed to determine whether members of the STAT family interact also with the δ-opioid receptor (δ-OR) and whether STAT5 phosphorylation is universal among the opioid receptors. Our results indicate that STAT5A and STAT5B interact with μ-OR and δ-OR respectively, but the strength of association between these interacting partners differs depending on the activation status of the opioid receptors. Opioid receptor stimulation leads to STAT phosphorylation by Src kinase and subsequently phosphorylated STATs activate gene transcription. Comparative analysis of the Gα proteins implicated in the STAT5A/B activation by μ-OR and δ-OR suggests that although endogenous Giα/Goα proteins are involved in δ-OR-activated STAT5Β phosphorylation, STAT5A stimulation by μ-OR is mediated by alternative mechanisms.
