Summary Hyalectans or lecticans (aggrecan, versican, neurocan and brevican) are large aggregating proteoglycans (PGs) that carry mainly chondroitin sulphate (CS) side chains. The brain contains also decorin and biglycan. The aim of this study was to evaluate the localization and the distribution of hyalectans and small PGs decorin and biglycan, in cerebrum (CB), cerebellum (CL) and brainstem (BS) of young sheep brain. Brain PGs were extracted from delipidated CB, CL, and BS and were analyzed by biochemical methods, including immunoblotting and high performance liquid chromatography (HPLC). Moreover, brain sections were examined immunohistochemically using a panel of antibodies directed against hyalectans. Biochemical analysis indicated that versican’s immunoreactivity was stronger in BS, in contrast to neurocan that was stronger in CL while aggrecan was greater in the CB and BS than the CL. Immunoreactivity for decorin and biglycan was of about the same density in CB, CL and BS. Disaccharide analysis in the three areas of the brain showed that Δdi-4S is the major component in the CS/DS chains of the brain PGs, while the ratio of Δdi-4S/Δdi-6S appears to be reduced in BS compared to CB and CL.
