Summary Previous study carried out on PC12 cells expressing α2 adrenergic receptors have shown that stimulation of all three subtypes causes morphological and molecular neuronal differentiation similar to that by growth factors, but the signaling mechanisms that mediate this effect have not been characterized. As an initial effort to delineate the signal transduction mechanisms employed by the α2-ARs for this effect, we investigated the involvement of Receptor tyrosine kinases and found tha the activation of MAPK and Akt by α2-ARs in PC12 cells requires the consecutive transactivation of two receptor tyrosine kinases, an early activation of EGFR and a late transactivation of TrkA. These results reflect the remark- able diversity of the signal transduction pathways utilized by a by a α2-ARs in order to elicit critical signaling events.
