Summary | This study gives information about the characteristics of the rabbit LES tone following in vitro application of electrical field stimulation (EFS). At frequencies 0.5, 1, 2, 4, 8, 16 and 32 Hz, EFS produces an on relaxation followed by a frequency-dependent off contraction. NO seems to be involved in both responses, since L-NAME influences the on relaxation either by reversing it to an on contraction (at frequencies from 0.5 to 2 Hz) or by reducing it (at 8, 16 and 32 Hz), while it decreases the off contraction at all frequencies tested. The involvement of purinergic transmitters in both responses is also suggested, as suramin blocks the non-nitrergic component of the on relaxation, while the P2Y1-antagonist MRS 2179 reverses it to an atropine- and TTX-sensitive on contraction suggesting the participation of a cholinergic transmitter uncovered after blockade of P2Y1-receptors. In the same line, the L-NAME-insensitive component of the off contraction is reduced by suramin and it is fully blocked by MRS 2179. Conclusively, results of this study suggest that both nitrergic and non-nitrergic neurotransmitters regulate the rabbit LES tone. | ||
