Summary | Adenosine A1 receptors (negatively coupled to G proteins) are co-localized and interact antagonistically with dopamine D1 receptors (positively coupled to G proteins) on striatal neurons, regulating through the cAMP/PKA/DARPP-32/PP-1 pathway the activity of the direct pathway of the basal ganglia. In the present study, we have demonstrated by using the in situ hybridization technique, that CPA (an adenosine A1 receptor agonist) when co-administrated with SKF38393 (a dopamine A1 receptor agonist) attenuates the 01-mediated zifI268 mRNA expression in striatum of the weaver mouse. Weaver mutation is a genetic model of dopaminergic deficiency. Unexpectantly CPA, when administrated alone, induces zif/268 mRNA expression in striatum and all layers of barrel field cortex of the weaver, but not of control mice. The aforementioned induction of zif/268 mRNA is not counteracted by co-administration of CPA and ZM241385 (an adenosine A2A receptor antagonist) confirming that the induction is A1 receptor mediated. However this up-regulation of zifI268 is significantly reduced in striatum by co- administration of CPA and Quinpirole (a dopamine D2 receptor agonist), indicating a probable A1/D2 receptor interaction. Furthermore, the A1-mediated zif/268 mRNA induction is counteracted by using U73122, an inhibitor of phospholipase C (PLC). In conclusion, (a) adenosine A1 receptor activation can counteract the dopamine D1 receptor mediated zif/268 mRNA expression in weaver striatum confirming the A1/D1 receptor interaction taking place on striatal neurons of the direct pathway, (b) the unexpected zif/268 mRNA up regulation elicited via A1 adenosine receptor stimulation, indicates a supersensitivity of these receptors, which could probably rely on a switch in A1 receptor function under dopamine deficiency, using probably the PLC signal transduction pathway. | |
