Περίληψη – Summary
|
– This study reveals differential dose-related effects and mechanisms of theophylline’s action along the rabbit digestive tract. Up to the concentration of 10-3 M, theophylline relaxes all regions of the upper gastrointestinal tract, in a declining order from the lower esophageal sphincter (LES) to the pylorus, but only the ascending colon from the lower gastrointestinal tract; this effect persists under non-adrenergic non-cholinergic (NANC) conditions and it is not modified by hexamethonium (HEX) or by L-NAME. At concentrations higher than 10-3 M, the theophylline’s relaxing effect on the antrum and pylorus is reversed into a considerable atropine-sensitive contractile one. In the small intestine, theophylline produces a weak atropine- and HEX-sensitive increase in tone, which at concentrations higher than 10-3 M becomes very strong. Available information suggests that the relaxing effect of theophylline is due to cAMP elevation mediated through its inhibitory action on the phosphodiesterases (PDEs), while its stimulatory effect involves a cholinergic pathway. |
