Summary Liposomes are composed of varying numbers of alternating concentric phospholipid bilayers and aqueous spaces. These vesicles are able to entrap water soluble or lipid soluble substances. Regarding their distribution in the body, liposomes are entraped by the reticuloendothelial system. This localisation of liposomes in circulating and fixed macrophages after i.v. injection offers a convenient method for “targeting” materials to these cells. The construction of liposomes bearing ligands that recognize molecular determinants on the surface of the target cells, in order to alter such natural localization, is also determined. Finally the liposome-drug preparations administered directly in the eye, the respiratory tract and the skin have an increased popularity and successful application. Drug delivery, using liposomes, to the mononuclear phagocyte-system, have already achieved practical reality, but several problems remain to be solved before the clinical use of liposomes.
