Summary | Three different brands of paracetamol tablets were evaluated for their mechanical strength, dissolution rate, and bioavailability. The in vivo evaluation of comparative bioavailability was studied in eight healthy volunteers using a cross-over study. The urine was collected for 24 hours and after hydrolysis of paracetamol and its conjugates the total drug was determined by the method of Welch and Conney. The relative bioavailabilities, with respect to a standard equivalent solution of 500 mg of paracetamol, were found to be statistically not significant. The dissolution rate was determined according to the method of USP XXI and the dissolution rate constants were found to be also statistically not significant. In contrast, the mechanical strength was found to differ significantly. This points out the inadequacy of mechanical strength to predict the dissolution rate of tablets. | |
