Summary Interactions between drugs and between drugs and excipients that influence absorption from the gastrointestinal tract into the systemic circulation are reviewed. This kind of pharmacokinetic drug interactions include in vitro drug interactions or interactions in the intestine. Interactions during the absorption phase may affect the amount of drug absorbed or the rate of absorption. Drug interactions have been classified as indirect, where the effect of one drug on gastrointestinal tract physiology influences the absorption of other drugs, or direct interactions involving pH changes, complexation, chelation, absorption and changes of the drug’s physicochemical properties (solubility, crystal form, particle size, etc.). Drugs may interact with excipients by a number of different mechanisms producing as a result changes in dissolution rates, equilibrium solubilities and chemical stability.
