Summary | The mechanism of action of two new 4-quinolones on whole rat brain changes of serotonin (5-HT) metabolism was examined. 12 groups of 90 male wistar rats 230±0g of weight were used. Each group received single i.p. injections of saline (control, C), ciprofloxacin (Ci), ofloxacin (O) each in 20 mg/kg, or a combination of pargyline (P) 75 mg/kg, reserpine (R) 5 mg/kg, parachlorophenylalanine (PCPA) 100 mg/kg, with saline or each quinolone in the same dose and a suitable treatment. Animals were killed 2 h after injection of serum or each quinolone and whole brain 5-HT and 5-HIAA were fluorometrically determined. A significant increase of 5-HT levels with no changes of 5-HIAA/5-HT ratio under both quinolones treatment was observed. This was not an anti-MAO effect since no reinforcement of (P) action on both 5-HT and 5-HIAA levels was noticed. Both quinolones reversed the action of (R), more significantly by (O), a finding which suggests that a membrane stabilizing effect is exerted and is (at least partly) involved on quinolones’ effect on 5-HT levels. A slight inhibitory effect of each quinolone upon PCPA action on 5-HT levels was observed. These findings do not provide firm conclusions of the exact mechanism that quinolones significantly influence brain 5-HT. Possible involvement of quinolones on tryptophan availability to brain or 5-HT interaction with other neurotransmitter pathways is a matter for further elucidation. | |
