Summary | Atrial natriuretic peptide (ANP) and its receptors have been found in cerebral areas (hypothalamus and brain stem) involved in blood pressure and heart rate (HR) regulation. However, the exact effects of intracerebroventricular (icv) administration of ANP on the circulatory system remain unresolved. To evaluate these effects mean blood pressure (MBP), HR, change in central venous pressure (dCVP) have been measured and the double product (DP) has been calculated in 10 New Zealand White (NZW) rabbits [Body Weight (BW) = 2.86 ± 0.39 kg, Hematocrit (Ht) = 37.55 ± 1.1 %] before and after the bleeding of 20% of blood volume (BV) [Rabbits BV(ml) = -47.587 + 197.342x Ln(BWkg)] . One week before bleeding, an indwelling icv cannula was implanted in the right lateral ventricle and two indwelling catheters, intracarotid and intrajugular, were inserted in the corresponding vessels. The rabbits were divided to two groups (A and B) of 5 animals each. The circulatory parameters were recorded before (0 min) and 10, 20, 30, 40, 50, 60 min after bleeding and the subsequent icv administration of either 25 μl artificial cerebrospinal fluid [aCSF] (group A) or 25 pi rANP (1 μg) in aCSF (group B). The results, analysed by repeated measures ANOVA and t-test, indicate a post bleeding longer lasting increase in HR (p<0.05) in group B (ANP receivers). These results suggest that in rabbits icv administration of rANP in physiological amounts (1 μg ~ 300 pmoles) alters the circulatory reaction to acute hypovolemia. This would indicate that brain ANP may participate in the cardiovascular system regulation under specific conditions. | |
