Summary La/SSB autoantigen is a ribonucleoprotein particle consisting of a protein component against which are often directed autoantibodies in sera of patients with primary Sjogren’s Syndrome (pSS) and Systemic Lupus Erythematosus (SLE). The purpose of the present study is to investigate the fine specificity of these autoantibodies by determining the B-cell epitopes of the La/SSB autoantigen. The epitope mapping of the La/SSB protein was performed using overlapping 20mer synthetic peptides covering the entire sequence (408 amino acids) of the protein. Purified IgG from sera of SLE and pSS patients were tested against these overlapping synthetic peptides. Peptide sequences spanning the regions 145-164, 289-308, 301-320 and 349-368 of the La/SSB protein were found highly reactive with the purified IgG. Biotinylated synthetic soluble peptides corresponding to the minimum required length of the epitopes were also prepared. The defined epitopes:

showed significant sensitivity and specificity for the anti-La/SSB antibodies of about 94 and 86% respectively.
