Summary Epigenetic regulation of gene expression via chromatin structural modification, including DNA methylation and histone acetylation status, has been considered as a key developmental process in higher mammals. In an attempt to uncover the role of both DNA and RNA methylation in the initiation of murine haematopoietic cell differentiation, we observed that treatment of MEL cells with inhibitors of methylation blocked the differentiation process and led to accumulation of discrete, relatively low molecular weight RNA transcripts hybridized with the 3’- flanking sequences of βmajor globin gene. Although the precise structure and origin of these RNAs as well as their role in the inhibition of MEL cell differentiation are not known, detailed analysis of the 3’-flanking DNA sequences of βmajor globin gene have revealed that it contains consensus sequences for the GATA-1, NF-E2 and EKLF transcription factors and a 81-repeat element. The central aim of the present study is to determine whether this area serves as a promoter-like region activated by hypomethylating agents through the binding of specific transcription factors in a way that may be implicated in the blockade of erythroid differentiation in vitro.
