Summary The purpose of this study was to elucidate the effect of central and peripheral deafferentation of spinal motoneurons in neonatal animals and to investigate the role of magnesium ions as an NMDA antagonist in order to prevent neuronal cell death. Sciatic nerve crush or spinal cord transection was performed in neonatal rats and the motoneurons in the lumbar cord were stained, detected and counted in the adult animals. Retrograde labeling with the HRP showed that lumbar motoneurons were not reduced in transected animals, whereas there was significant reduction in the axotomized ones. Administration of magnesium resulted in increased motoneuron survival compared to the non-treated (normal saline injected) animals. We conclude that in neonatal life neurons do not depend so much on their central connections for their survival, but, on the other hand, axotomy provokes considerable motoneuron loss, which can be avoided by administration of NMDA antagonists.
