Summary | Following the isolation of the guinea pig motilin (gp mo), we studied its potency, in comparison with the porcine one, in the rabbit duodenum, and its bio-activity in both unstimulated and stimulated guinea pig antrum and intestinal strips. In the rabbit duodenum the pED50 values for gp and porcine motilin were 8.35 and 8.79, respectively. In the guinea pig, the responses to gp mo were rather weak, since a high concentration was needed to obtain them. 70% of the unstimulated segments responded to 10-6 M gp mo, with jejunum and ileum to be the most sensitive and to give a mean response of 10% of the response to 10-4 M Ach, reaching in the ileum a maximum of 30%; the mean responses of the duodenum and the ascending colon were less than 5%, reaching a maximum close to 10%. This effect was antagonized by atropine and tetrodotoxin (TTX), and, as far as the small intestine is concerned, by the 5HT4-antagonist GR-113808. The reproducibility of the data was very small in the stimulated strips, since only 30% of them responded to gp mo. A shift in the base line in the longitudinal strips and a potentiation of responses to electrical field stimulation (EFS) in the circular strips of the duodenum and ileum were observed. In conclusion, guinea pig and porcine motilin exhibit comparable potencies in the rabbit. In the guinea pig, the weak responses obtained for gp mo seem to be both muscle- and neurally-mediated. | ||
