Summary The effect of aging in high affinity dopaminergic binding in the brain of Long-Evans rats was studied by using the specific D2-like receptor antagonist [³H]spiperone. Our data showed a statistically significant decrease in high affinity [³H]spiperone binding in hippo- campus, cerebellum, cerebral cortex and striatum of the aged (24 months), compared to young (3 months) rats. However, no changes in high affinity [³H]spiperone specific binding was observed in the midbrain and pons medulla. The above decreases reflect a decline in maximal density of D2-like receptor binding sites rather, than a change in the Kd value, since the Bmax values were significantly smaller compared to the young animals. The decreases of the D2-like dopamine receptors, seen in hippocampus, cerebellum, cerebral cortex and striatum, indicate a brain area specific decline of the dopaminergic system function during aging.
