Summary | The present study examines the effect of the widely used bronchodilator drug theophylline, a non-specific phosphodiesterase (PDE) inhibitor, on the rabbit oesophagus and lower oesophageal sphincter (LOS) contractility, and explores the mechanism of its action. According to the results theophylline, at concentrations higher than 3×10-5 M produces a concentration-dependent decrease in the tone and/or the phasic contractions of the rabbit oesophagus and LOS. This effect is not antagonised by adrenergic and cholinergic antagonists or by the ganglion-blocker hexamethonium (HEX), suggesting that it is non-adrenergic, non-cholinergic (NANG) and post-synaptic. In addition, L-NAME, an inhibitor of nitric oxide synthetase, fails to modify the theophylline’s relaxing effect. In conclusion, theophylline, at quite high concentrations, produces a post-synaptic, concentration-dependent NANG decrease in the tone and/or the phasic contractions of the rabbit oesophagus and LOS. There is evidence to suggest that this relaxing effect is due to cAMP elevation mediated through the inhibitory action of theophylline on the PDEs present in the LOS and oesophagus tissues. | |
