Summary | In the present study, the effect of theophylline on the phasic motor activity and the basal tone of the rabbit duodenum, jejunum, ileum and ascending colon were investigated. In all three different parts of the small intestine, theophylline, at concentrations ranging from 10-6 to 10-3 M, produces an atropine-sensitive increase in the amplitude of phasic contractions, while it does not significantly influence the basal tone. In the ascending colon, theophylline, at concentrations ranging from 10-7 to 3×10-5 M, does not affect the phasic contractions, whereas it produces an atropine-sensitive increase in the basal tone; at concentrations higher than 3×10-5 M, theophylline reduces the phasic contractions and it produces a sustained decrease in the basal tone, an effect that is not influenced by atropine. In conclusion, the theophylline’s effect on the rabbit intestine varies depending upon both the intestinal region and the drug’s concentration. Thus, theophylline either stimulates the intestinal motility by potentiating the phasic contractions in all three parts of the small intestine and by increasing the basal tone in the ascending colon, probably via a cholinergic pathway, or it inhibits the intestinal motility by producing a non-cholinergic sustained decrease in both the phasic contractions and the basal tone of the ascending colon. | |
