Summary | TGF-β1 is included among main cytokines involved in the regulation of pathogenesis of rheumatoid arthritis and osteoarthritis, and is considered that it plays a protective role. TGF-β1 (1ng/ml) significantly enhances the production of IL- 6 and TIMP-1 by human synovial fibroblast in a dose (0.01-1ng/ml) and time (12-48h incubation) dependent manner. Using specific protein kinases inhibitors, it is shown that this process is mediated by protein kinase C. Prothymosin a, a bioactive acid peptide, detected in synovial membrane and fluid, dose-dependently (0.1-1pg/ml) suppresses the TGF-β1 -enhanced production of TIMP-1 but not of IL-6 by fibroblasts. The role of prothymosin a in arthritis may be considered as pro-inflammatory. | |
