|
– Fentanyl, an m-opioid receptor agonist, has long been studied for its neuropharmacological effects. However, its action on the release of neurotransmitters is not totally understood. The results from studies on the cardiovascular changes after fentanyl administration are conflicting. In the present study the action of fentanyl on the concentration of the excitatory and inhibitory aminoacids is investigated. We examined the effects of continuous intravenous (10 μg/kg) as well as intrahypothalamic (50 nmol/min) fentanyl administration on the release rates of glutamate in the superfusate of hypothalamus, under tail pinch manipulation. The release of the neurotransmitter was monitored by the push–pull superfusion technique. To investigate the role of fentayl we also administered an opioid antagonist, naloxone 0.01 mg/kg iv, or 0.1 nmol/min intrahypothalamicaly. Glutamate was determined in the above solution by High Performance Liquid Chromatography (HPLC) and fluorimetric detection after OPA derivatisation. After intravenous fentanyl administration a significant decrease of glutamate was observed. However during the pain manipulations the release rates of glutamate was increased. Intravenous and intrahypothalamic fentanyl administration caused fluctuations in blood pressure and heart rate. Naloxone iv failed to reverse the cardiovascular as well as neurological effects, while intrahypothalamic antagonist administration reversed the alterations neurotransmitter release rate. We conclude that fentanyl effects are related with glutaminergic system activation. There is a possibility that the changes of the amino acid concentration in hypothalamus are related with the parasympathetic inhibition after continuous fentanyl administration. |
