Έχει διαπιστωθεί ότι οι ουσίες που προκαλούν εξάρτηση προκαλούν μεταβολές στις νευροχημικές λειτουργίες με την έννοια ότι μιμούνται ή αυξάνουν τις ενέργειες των φυσιολογικών νευρομεταβιβαστών. Οι περισσότερες από τις ουσίες αυτές επιδρούν σε κάποιο υποδοχέα ή μεταφορικό σύστημα. Τα κυκλώματα του εγκεφάλου που είναι υπεύθυνα για την ανάπτυξη του εθισμού έχουν αρχίσει να γίνονται κατανοητά περισσότερο στα πειραματόζωα. Το κυριότερο από τα κυκλώματα αυτά φαίνεται να είναι εκείνο της δοπαμίνης, που αρχίζει από την κοιλιακή μοίρα της καλύπτρας των εγκεφαλικών σκελών και διά μέσου του λοφιδίου του κερκοφόρου πυρήνα φθάνει στον προμετωπιαίο φλοιό. Υπάρχουν τουλάχιστον 80 διαφορετικοί νευρομεταβιβαστές στον εγκέφαλο και ορισμένοι από αυτούς φαίνεται ότι έχουν σχέση με τον εθισμό. Drugs that provoke dependence produce alterations in brain function by initiating neurochemical processes that usually mimic or increase the action of endogenous transmitter. Most of these drugs have an action on a certain type of receptor or on a transporter site. Brain circuits of addiction are beginning to be understood more in animals. The primary circuit seems to be the dopamine pathway that runs from the ventral tegmental through the nucleus accubens to the profrontal cortex. There are at least 80 other brain neurotransmitters some of which are likely to be involved in addiction. |
