In the present study, we demonstrate that activation of the δ-opioid receptor (δ-OR) with either the δ specific opioid agonist DSLET or morphine leads to STAT5Β tyrosine phosphorylation in HEK293 cells stably transfected with the δ-opioid receptor and endogenously expressing STAT5B. Moreover, we demonstrate, by pull-down experimentation, employing a fusion peptide encompassing the C-terminal region of the δ-opioid receptor, that STAT5Β interacts directly with the C-terminal tail of the δ-opioid receptor. On the other hand, using a truncated version of the C-terminal tail of δ-opioid receptor that lacks the first 26 amino acids indicated that STAT5Β interacts within helix 8 of δ-opioid receptor. Immunoprecipitation studies showed that STAT5Β co-precipitates with δ-OR, confirming the interaction of STAT5Β with the receptor. Additional coimmunoprecipitation experiments indicated the direct binding of Gβγ subunits to δ-OR and STAT5B. Collec­tively, our data demonstrate that the ternary functional complexes formed between δ-OR, Gβγ and STAT5B after δ-OR activation mediate STAT5Β phosphoryla­tion.
