Περίληψη – Summary

Quinolones are one of the main antimicrobial groups used for the treatment of different infections. The success of their therapeutic outcome depends οn the wide antimicrobial spectrum, lipophilicity, pKa and penetration in tissues and biological fluids. The aim of the present study was to evaluate and compare the distribution of four quinolones (Norfloxacin, Ofloxacin, Ciprofloxacin, Pefloxacin) in samples of serum and saliva (both mixed and parotid) of 6 normal volunteers, with and without the co-administration of acetazolamide (carbonic anhydrase inhibitor). Drug administration took place in two phases: a) a single dose of each quinolone was administered p.o. between 6.30-8.30 am and b) acetazolamide was administered in 3 doses/6hrs and the last dose was given simultaneously with quinolones, as stated above. Blood and saliva samples were taken at 2 and 5 hours post-drug administration. Mixed saliva samples were obtained from the bottom of the oral cavity while parotid saliva collection was performed by means of the Lasley instrument. Quinolone levels in serum and saliva were measured by biological agar diffusion method with holes. Fluoroquinolones were detected in both saliva and serum with their concentrations peaking in serum and following the order: serum >parotid saliva >mixed saliva. Quinolone concentrations in saliva were proportional to those in serum, with their relevant concentrations following the pattern: Pefloxacin >Ofloxacin>Norfloxacin>Ciprofloxacin.Acetazolamide co-administration resulted in reduction of pefloxacin levels in serum, reduction of pefloxacin levels in parotid saliva and elevation of ciprofloxacin levels in both mixed and parotid saliva. Paired t-test was used for the statistical analysis.
