Περίληψη – Summary Although covering a wide spectrum of chemical properties, chemical carcinogens act in vivo, either directly or following metabolic activation, as electrophilic species, forming covalent adducts with cellular macromolecules. The formation of specific adducts with DNA is believed to be one of the first, critical steps in the process of carcinogenesis. During recent years significant progress has been made in the study of these complexes as well as in the methodology for their detection and quantitation, a development which offers new possibilities in the study of the aetiology of human cancer. There has also been increased appreciation of the importance of DNA repair in the protection of the ceil from the effects of carcinogenic stimuli. If a carcinogen-damaged cell does not repair its DNA in time before it undergoes a cycle of DNA replication, it may enter irreversibly a precarcinogenic stage, due perhaps to a change in the base sequence of the DNA. This first stage of carcinogenesis (initiation) may be followed by one or more stages (promotion) leading to the full neoplastic transformation of the cell.
