5-HT2C receptor is one of fourteen 5-HT receptor subtypes expressed in the CNS and iIt’s gene is located in the X-chromosome (Xq24). Both pharmacological and genetic studies reveal that it contributes substantially to the serotonergic regulation of a wide variety of behavioral and physiological processes such as feeding and glucose homeostasis. The polymorphism -759C/T in the serotonin 2C (5HT2C) receptor promoter region has been associated with the development of type II diabetes mellitus and obesity in normal subjects, with the frequency of the T allele being higher in subjects without diabetes mellitus and obesity. Furthermore, the 5-HT2C-receptor gene -759C/T polymorphism has been associated with antipsychotic agents-induced weight gain. The T allele may be a protective factor for the development of weight gain in schizophrenic patients. Objective: To determine the distribution of the -759C/T polymorphism of the 5HT2C receptor among normal controls, in patients with type ΙI diabetes mellitus and in patients with coronary artery disease in the Greek population. Methods: Ιn this study participated 50 healthy volunteers, 50 patients with type ΙI diabetes mellitus and 50 with coronary artery disease. The experimental process included the extraction of genomic DNA from peripheral blood and then its analysis for the -759C/T polymorphism of the 5-HT2C receptor using PCR-RFLP method. Results – Discussion: In the group of healthy volunteers the frequency of T-allele in normal controls (19.2%) is in accordance to that reported in European populations and in patients with type II diabetes mellitus and in patients with coronary artery disease is 8.5% and 17.7%, respectively. |
